Excipients and buffers: what else is in a lyophilized vial

AuraVia Research Team4 Min. Lesezeit

A lyophilized peptide vial rarely contains pure peptide alone. Most formulations include small amounts of other ingredients — collectively called excipients — that exist to make the freeze-drying process work and the resulting powder easier to handle and reconstitute predictably.

Why excipients are there at all

Freeze-drying a small mass of pure peptide on its own tends to produce a fragile, hard-to-handle residue rather than a clean cake or powder. Bulking agents — commonly sugars or sugar alcohols such as mannitol — are added to give the lyophilized product structure and make it easier to see, weigh conceptually, and dissolve consistently.

Buffer salts are often included as well, to stabilise the solution's pH during and after reconstitution, since a peptide's stability can be sensitive to pH drift.

What it means for concentration maths

Because a vial's total lyophilized mass includes excipients as well as peptide, the number that matters for concentration calculations is the peptide content stated on the label or CoA, not the visual size of the powder cake — a small-looking cake can still correspond to the labelled peptide mass once bulking agents are accounted for.

This is also why two vials with visually different amounts of powder can both be correctly labelled at the same peptide mass — the excipient load, not the peptide content, is what's driving the visual difference.

Reading a formulation note

When a product listing or CoA specifies a formulation (for example, a mannitol or buffer component), it's describing what's added alongside the peptide, not diluting the stated peptide mass — the labelled mg figure still refers to peptide content specifically. Checking this before reconstituting avoids a common source of concentration miscalculation.
